摘要:采用SSR技术,从105对柑桔SSR引物中,筛选出10对在柑桔大类品种间多态性位点丰富、带型清晰、重复性好的SSR引物。利用其中5对SSR引物对5个柚和7个杂柑品种DNA进行PCR分析,共扩增出35条谱带,其中27条谱带呈现多态性,引物多态性带纹比例均在60%以上。扩增片断的大小集中在100bp~500bp之间。分别针对柚和杂柑品种筛选出两个高效的引物组合(SSR17、SSR15、SSR16、SSR12与SSR17、SSR18、SSR16),利用这两组引物成功构建出了我国主推的5个柚和7个杂柑品种的特征指纹图谱。并应用指纹图谱自动识别软件Gel2.0对各品种指纹图谱进行识别,建立了相应品种的指纹模式图谱库。

Title: Identification of Pummelo (Citrus grandis (L.) Osbeck) and Hybrid Citrus Cultivars using SSR Molecular Markers

Abstract: Simple sequence repeat(SSR)was used to construct the fingerprinting of mainly cultivars of pummelo(Citrus grandis(L.)Osbeck)and hybrid citrus in China.A total of ten primer pairs with stable and repeatable polymorphism that can identify closely related citrus cultivars were screened out from 105 SSR primer pairs.In this paper,five pummelo and seven hybrid citrus cultivars were analyzed by using five of these SSR markers.As a result,a total of 35 bands were detected,27 of which were polymorphic.The polymorphic rate of each primer pair was beyond 60%.The bands range in size was from 100 bp to 500 bp.The fingerprinting of five pummelo and seven hybrid citrus cultivars was constructed by using two particularly high efficiency primer groups(SSR17、SSR15、SSR16、SSR12 and SSR17、SSR18、SSR16)respectively.Furthermore,the mode fingerprinting datebase of citrus cultivars was established by fingerprint analysis system-Gel 2.0.
