摘要：利用改进的CTAB法提取了15个柑橘分类群的基因组DNA,并进行了AFLP分析。结果表明,该方法提取的DNA溶液无色透明,紫外分光光度计测得A260/A280为1.8~2.0,1%琼脂糖胶电泳为清晰的一条带,RNA去除干净,无降解现象。其AFLP分析(引物EcoRI-AAC+MseI-CAC)条带清晰,多态性好,说明此方法完全适于AFLP分析。

Title: DNA Extraction Method for AFLP Analysis in Citrus

Abstract: The Genomic DNA samples of 15 taxa citrus were isolated by modified CTAB method and then AFLP analysis was conducted. The result indicated that the extracted DNA by this method appeared as a clear band, and when electrophoresed in 1% agrarose gel the RNA was eliminated completely. Measured the DNA by smartspec TM 3 000,the value of A260/A280 was 1.80~2.0. AFLP analysis indicated the primer (EcoRI-AAC+MseI-CAC)produced clear polymorphic patterns. Therefore this method could be used for extracting ideal DNA samples and be suitable for AFLP.
