摘要:Citrus is an important commodity worldwide and produced in tropical and subtropical regions around the world. Genetic transformation of Citrus via Agrobacterium is a valuable technique for citrus improvement due to difficulties of conventional citrus breeding. In the present study we carried out genetic transformation studies through Agrobacterium rhizogenes ATCC15834 including RolABCD genes to investigate the effects of different acetosyringone concentrations on transformation assays. In the study; stems and leaves of Gou Tou sour orange, Tuzcu 3131 sour orange and Carrizo Citrange were used as explant sources. All explants were obtained from germinated seeds in in vitro conditions. 0, 25, 50, 100 and 200 μM acetosyringone concentrations were used with all explants in transformation studies. Shoots and roots proliferations were scored at 4-8th weeks after transformation. All data was calculated as percentage and arcsine-transformed. Means were separated by analysis of variance and the LSD test was performed to examine significant differences in shoot and root formations coming from stem and leaf explant types at different acetosyringone concentrations. As a result; the best shoot formations were detected at stem explants and 25 μM acetosyringone for Gou Tou, stem explants and 100 μM acetosyringone for Carrizo, stem explants and 200 μM acetosyringone for Tuzcu 31 31. The best root formations were detected at leaf explants and 25 μM acetosyringone for Gou Tou, stem explants and 50 μM acetosyringone for Carrizo, leaf explants and 25 μM acetosyringone for Tuzcu 31 31 sour orange rootstocks.
