摘要:By exploring factors which had influences during AFT gene transfer to citrus,transformation system of epicotyls from C.paradisi seedling epicotyls were established and transgenic plant were obtained in this paper.The transformation frequency was assessed in response to several different factors,including antibiotics type concentration,pre-culture time medium,co-cultivation media,dark incubation in on sets of selection.Among the antibiotics,timentin was better than cefotaxime sodium Ceftriaxone sodium for differentiation of adventitious buds growth control of agrobacterium.3days of explants pre-culture in pre-culture medium plus 0.75mg/L TDZ,co-culture in medium adding 0.28mg/L DTT and 5%trehalase,and growing in darkness for 5-7days after co-culture were help to Kan anti-bud developing.Transgenic plants were obtained by PCR assay.
