标题:In planta genome editing in citrus facilitated by co‐\expression of CRISPR/Cas and developmental regulators

摘要:Recent advances in genome editing offer transformative potential for fruit tree breeding, yet conventional methods relying on tissue culture remain laborious and prone to somaclonal variation. Here, we present *in planta* genome editing in citrus (IPGEC), a novel Agrobacterium-mediated system enabling direct genome editing in soil-grown seedlings without tissue culture. IPGEC integrates transient co-expression of three functional modules: (1) CRISPR/Cas9 machinery targeting *CsPDS* (a carotenoid biosynthesis gene), (2) developmental regulators (e.g., *WUS*, *STM*, *IPT*) to enhance *de novo* shoot regeneration, and (3) T-DNA delivery boosters (VIP1/VirE2) under a cell division-specific promoter (YAO). This approach achieved up to 75% regeneration efficiency in Carrizo citrange, with multiplex editing confirmed by albino phenotypes and sequencing. Notably, high-throughput screening identified transgene-free chimeric or fully edited shoots (e.g., line #81), demonstrating IPGEC’s potential for non-GMO breeding. Developmental regulator efficacy varied across cultivars (Carrizo, Duncan, Foster, Hudson), with IPT or GRF4-GIF1 optimizing transformation rates (12.5–30%). By bypassing tissue culture and enabling transient editing, IPGEC accelerates trait-specific improvements in elite citrus varieties while mitigating somaclonal risks. This system establishes a framework for precision breeding in perennial crops, addressing challenges of heterozygosity and long generation cycles in fruit trees.
