标题:Allelic pattern of SSRs with high hybrid detection efficiency and inheritance of leaf petiole wing in interspecific Citrus crosses

摘要:Introduction - SSR markers can differentiate zygotic hybrids from nucellar seedlings in Citrus crosses, but with a variable hybrid detection efficiency. The use of SSRs with different polymorphism patterns may elucidate the cause of this variability. Petiole wing is an important morphological trait to differentiate the cultivated Citrus groups, but its genetics is not clear. The progeny of heterozygous individuals may show segregation in F, generation itself and thus, useful for studying the inheritance of traits. Materials and methods - In this study, hybrids were developed among three Citrus species namely, Jatti Khatti (JK), a strain of rough lemon (Citrus jarnbhiri Lush.), Rangpur lime (RL) (C limonia) and Sour orange (SO) (C. aurantium) by crossing JK x SO, RL x SO, and SO x RL. The parents were assayed with 30 SSR markers to identify the polymorphic SSRs for hybrid identification. The parents had differences in the length and width of petiole wing. To infer the inheritance pattern of petiole wing, the data of length and width of petiole wing were recorded for parents and the hybrid population, and was checked for its normality distribution. Results and discussion - From the polymorphic SSRs, five SSRs (CATOI, CT21, Ci08C05, CiBE4559, and CMS14) were used for hybrid identification which represented three different polymorphic patterns: homo-allelic polymorphic (HAP), maternally hetero-allelic, polymorphic (MHAP) and paternally hetero-allelic, polymorphic (PHAP) in the parents. The HAP-SSRs proved to be least informative and identified least number of hybrids with HDE ranging from 54.54 to 66.67%. The PHAP-SSRs that amplified two polymorphic alleles in male parent, unambiguously detected all the hybrids in all the crosses. The HDE of the markers was influenced by the occurrence and frequency of null alleles of male parent. The Shapiro-Wilk's test revealed that petiole wing length in the F, progenies of JKx SO (p = 0.090), RL x SO (p = 0.510), and SO x RL (p = 0.469) and in the combined F, progeny of RL x SO and SO x RL crosses (p = 0.324) followed normal distribution. Conclusion - The PHAP-SSRs should prove most informative for hybrid detection in Citrus. The normality of the petiole wing length indicates its governance by polygenes.
