标题:A citrus pulp mastication trait-related ABSCISIC ACID-INSENSITIVE 5-like protein promotes fruit softening via transcriptionally activating pectate lyase gene

摘要:Mastication trait of citrus fruit is an important criterion to evaluate mandarin citrus fruit quality and decides its market value. However, the regulatory mechanism underlying the mastication trait formation is unclear. Here we isolated a bZIP family member, annotated to be "ABSCISIC ACID-INSENSITIVE 5-like protein 5", from fruits of two mandarin citrus cultivars with different mastication trait, and named it CsABI5-L. Amino acid sequence analysis was performed to confirm the conserved motifs in encoding protein sequence of CsABI5-L. Phylogenetic analysis and subcellular localization assay further indicated that it was a transcription factor belonged to group A of bZIP family. Heterologous expression of CsABI5-L in tomato resulted that fruit firmness of mature transgenic fruits were decreased compared with the wild type. Meanwhile, vertical and horizontal sections showed that the mesocarp and placenta of transgenic fruit were thinner than those of wild type. A transcript level analysis indicated that the expression of tomato cell wall degradation-related genes SlPG2, SlPE1, SlPL, SlTBG4 and SlEXP increased. A dual-luciferase transient expression assay demonstrated a specific binding of CsABI5-L to SlPL promoter, which was the putative reason causing the decrease in fruit firmness of transgenic lines. Our findings provide insights into the transcriptional regulatory mechanism controlling the mandarin citrus mastication trait.
