摘要:以自橙汁中分离的酵母菌株为材料,从培养与制备条件出发,研究不同的培养基、培养容器、培养时间、菌液量和菌液浓度对酵母菌基因组DNA提取质量的影响,建立了酵母菌基因组DNA优良提取体系.即以YPD为液体培养基,三角瓶(100 mL)为培养容器,在30℃下震荡培养12~16 h至菌液OD600=1.5左右,然后用此菌液10 mL以上按试剂盒提供的方法提取基因组DNA.电泳结果表明,采用该体系提取的酵母菌基因组DNA量大、质量好.

Title: Effects of Culture and Preparation Conditions on Extraction Quality of Genome DNA from Yeast

Abstract: Thirty yeast strains were separated from orange juice and used as the experimental materials.Based on the culture and preparation conditions,the effects of different culture vessels,culture time,yeast-liquid volume and yeast-liquid optical density on the extraction quality of genome DNA from yeast were studied,and an improved yeast genome extraction system was set up successfully.The yeast strains were cultured in YPD liquid medium enclosed in conical flasks at 30 ℃ for 12~16 hours until the OD600 of the yeast cultural liquid was close to 1.5.More than 10 mL of the yeast liquid was taken for extracting the genomic DNA according to the procedures of the kit.The results of electrophoresis showed that both the mass and quality of the genomic DNA harvested were highly satisfactory.
